It is a bit far from Rio or Sao Paulo, it will closer to Sao Luis in the North. It is complet worthwile.
You can through Barreirinhas, it is a gateway to the park, but if you go to Sao Luis *Maranhao State and them find some good agency is cheaper them going straight to Barreirinhas.
I assure you will have fun, specially if you are more connected to nature and last touristic city...
